I'd think some sort of straight "stick" in the muzzle would quickly show any misalignment. In fact, that's what the instructions say to do when it's mounted on the barrel to check that it's parallel with the bore and not too close or too far from the bullet's path.

Mine was probably due to an enlarged primer pocket but it was too long ago to remember. It went off about the same time the bullet dislodged and the powder dumped and there was no powder burned. But if it had, I doubt it would have blown up as the hammer's not exactly sealed. Watch out...
It's my understanding there's not a result that can be termed 100% negative because an animal can be positive with prion levels not high enough to be detected yet. Rather, the results are either positive or not detected.
Soaking in 50:50 bleach solution for an hour is cited as working. From what I've seen of even a 10% solution, it's fairly corrosive though. You could freeze quarters and process after getting results.
